=== Intake of glucose by mouth === The blood glucose can usually be raised to normal within minutes with 15–20 grams of carbohydrate, although overtreatment should be avoided if at all possible. It can be taken as food or drink if the person is conscious and able to swallow. This amount of carbohydrate is contained in about 3–4 ounces (100–120 mL) of orange, apple, or grape juice, about 4–5 ounces (120–150 mL) of regular (non-diet) soda, about one slice of bread, about 4 crackers, or about 1 serving of most starchy foods. Starch is quickly digested to glucose, but adding fat or protein retards digestion. Composition of the treatment should be considered, as fruit juice is typically higher in fructose which takes the body longer to metabolize than simple dextrose alone. Following treatment, symptoms should begin to improve within 5 to 10 minutes, although full recovery may take 10–20 minutes. Overtreatment does not speed recovery, and will simply produce hyperglycemia afterwards, which ultimately will need to be corrected. On the other hand, since the excess of insulin over the amount required to normalize blood sugar may continue to reduce blood sugar levels after treatment has produced an initial normalization, continued monitoring is required to determine if further treatment is necessary.

=== Nutritional supplements === Supplementary "protein substitute" formulas are typically prescribed for people with PKU (starting in infancy) to provide the amino acids and other necessary nutrients that would otherwise be lacking in a low-phenylalanine diet. Tyrosine, which is normally derived from phenylalanine and which is necessary for normal brain function, is usually supplemented. Consumption of the protein substitute formulas can actually reduce phenylalanine levels, probably because it stops the process of protein catabolism from releasing Phe stored in the muscles and other tissues into the blood. Many PKU patients have their highest Phe levels after a period of fasting (such as overnight) because fasting triggers catabolism. A diet that is low in phenylalanine but does not include protein substitutes may also fail to lower blood Phe levels, since a nutritionally insufficient diet may also trigger catabolism. For all these reasons, the prescription formula is an important part of the treatment for patients with classic PKU. Evidence supports dietary supplementation with large neutral amino acids (LNAAs). The LNAAs (e.g. Leu, Tyr, Trp, Met, His, Ile, Val, Thr) may compete with Phe for specific carrier proteins that transport LNAAs across the intestinal mucosa into the blood and across the blood–brain barrier into the brain. Its use is limited in the US due to the cost but is available in most countries as part of a low protein / PHE diet to replace missing nutrients.

== Mechanism by which ceramide signaling occurs == Currently, the means by which ceramide acts as a signaling molecule are not clear. One hypothesis is that ceramide generated in the plasma membrane enhances membrane rigidity and stabilizes smaller lipid platforms known as lipid rafts, allowing them to serve as platforms for signalling molecules. Moreover, as rafts on one leaflet of the membrane can induce localized changes in the other leaflet of the bilayer, they can potentially serve as the link between signals from outside the cell to signals to be generated within the cell. Ceramide has also been shown to form organized large channels traversing the mitochondrial outer membrane. This leads to the egress of proteins from the intermembrane space.

=== Alloplastic grafts === Alloplastic grafts may be made from hydroxyapatite, a naturally occurring mineral that is also the main mineral component of bone. They may be made from bioactive glass. Hydroxylapatite is a synthetic bone graft, which is the most used now among other synthetic due to its osteoconduction, hardness and acceptability by bone. Tricalcium phosphate which is now used in combination with hydroxylapatite thus give both effect osteoconduction and resorbability. Polymers such as some microporous grades of PMMA and various other acrylates (such as polyhydroxylethylmethacrylate aka PHEMA), coated with calcium hydroxide for adhesion, are also used as alloplastic grafts for their inhibition of infection and their mechanical resilience and biocompatibility. Calcifying marine algae such as Corallina officinalis have a fluorohydroxyapatitic composition whose structure is similar to human bone and offers gradual resorption, thus it is treated and standardized as "FHA (Fluoro-hydroxy-apatitic) biomaterial" alloplastic bone grafts.
== Pharmacology == Computer models suggest fentanyl binds to μ-opioid receptors in its protonated form. Fluorination of fentanyl at the 3-position lowers the pKa of its conjugate acid from 8.4 to 6.8, a change designed to reduce its systemic effects while maintaining its potency in inflamed tissue, as this is a more acidic environment (pH 5–7) than non-inflamed tissue (pH 7.4). In experiments on rats with different types of inflammatory pain, it has been shown that NFEPP produces injury-restricted analgesia, while exhibiting reduced typical opiate effects such as respiratory depression, sedation, constipation, and chemical seeking behavior. As a result, NFEPP has the potential to reduce opioid addiction and dependency, as there is no effect on users who are not actually suffering from pain, as the chemical does not interact with non-inflamed brain tissue until much higher doses are reached. Further research indicates other fluorinated derivatives of fentanyl, such as β-fluorofentanyl (FF3) or 2'-fluoro β-fluorofentanyl (RR-49), may be more effective in achieving high potency in damaged tissue with low potency in undamaged tissue.
